Magnetic grapheneoxide(GO)–hemin probescontainingdisulfide bondsaresimplyandeffectively
synthesized throughamidereactiontocovalentlylinkmagneticparticlestoGOsurfaceand π–π stacking
interactionbetweenheminandGOtoimmobilizeheminonGO.Basedonthestrongnucleophilicityof
sulfhydry,wehavedevelopedacolorimetricdetectionsystemforthiolsbyusingglutathione(GSH)asa
model analyte.UpontheintroductionofGSHtothefabricatedmagneticparticle(MP)-GO–hemin probes,
the disulfides canbereadilyreducedbythiols,resultinginthereleaseofGO–hemin hybridstosolution.
Due totheexistenceofheminonGOsurface,thereleasedGO–hemin thathastheintrinsicperoxidase-
like activitycancatalyzetheoxidationofABTS2 by H2O2 to formthecoloredradicalproductABTS .
A broadlineardynamicrangeof1010M to106 M GSHisachievedwithadetectionlimitof
8.21011M (3s). Moreover,thenewprobeissuccessfullyappliedtothedetectionofnon-protein
thiols andproteinthiolsintheextractsofRamoscells,whichshowsfavorablecorrelationshipwiththe
results obtainedbyelectrochemicalmethod.Inaddition,theMP-GO–hemin probecandetectnon-
proteinthiolsinRamosextractsaslowas500cells.Inthisassay,thepreparedMP-GO–hemin conjugates
are thoroughlycharacterizedbySEM,AFM,UV–Vis, FT-IR,andRamanspectroscopy.
